    Different Types of DNA

    Autosomal STR: STRs are short tandem repeats found in the genetic material (or DNA) that makes up the human genome. Several different STRs, regions that are highly variable in length from individual to individual, have been located and examined. The size of the STR is what scientists look at to help answer questions regarding relatedness. The different sizes of DNA found at these STR regions correspond to, what geneticist call, alleles. These alleles are passed on from parents to offspring. By examining random individuals within a particular population, each allele is found to have a frequency associated with it within that group. It is these frequencies that researchers use to calculate the probabilities of relatedness. GeneTree uses STRs to measure probabilities of paternity, and most other DNA relationship test, including the Y-chromosome Haplotyping.

    How DNA is Analyzed

    To determine sizes of DNA in a particular region the DNA region is either:

    amplified using DNA primers, targeted to the regions, and a DNA polymerase enzyme (PCR method), or
    cut with restriction enzymes (RFLP method).

    The DNA samples are then ran through a system of capillaries using a chromotography method (called capillary electrophoresis) to resolve sizes. An example of an older procedure called gel electrophoesis is shown in Figure 1 to assist in illustrating the separation effect of chromotography. A computer-generated electropherogram (Figure 2) demonstrates how the DNA sizes can be obtained using a fluorescent detection system. It is similar to reading a barcode. This procedure has replaced the older gel electrophoesis method illustrated in Figure 1.

    To visualize the DNA, either:

    the primers are labeled before the PCR reaction (PCR method), or
    a labeled probe is used to hybridize to the specific region of DNA being examined (RFLP method).
    Once the sizes of DNA in the specific regions are determined, they are represented by a universal number (e.g., 9, 10, 11), and referred to as alleles.
    Each person should have 2 alleles at each system examined for paternity testing (with exception given to either, a) a person having the same allele presented twice, or b) the possibility of a ’silent’ allele). One allele is passed from the biological father and the other allele is passed from the biological mother.

    Each known allele has a determined frequency of occurrence in the general population and among different ethnic groups (Table 1). These frequencies are used to calculate the probabilities of paternity, siblingship, grand-parentage, etc.

    The mtDNA markers are maternally linked. They are passed from the mother to all of her children. Then, only the female children pass it on to all their children, and etc.

    Race or ethnic origins are common terms used to differentiate biogeographical population groups. While the notion of race or ethnic origin can be complicated and controversial, the most widely used categories are based on visible traits and self-identification. This is best illustrated by the FBI’s practical utilization of race to characterize the general appearance of suspects.

    The ability to infer a suspect’s race or ethnic origin from a DNA sample in a no suspect case can help prioritize an investigation.

    Ancestral Lineage Markers

    There are two principal types of ancestral lineage markers, paternal and maternal. Paternal lineage markers are located on the Y chromosome while maternal lineage markers are found in mitochondrial DNA (mtDNA).

    Ancestral lineage markers are passed down from generation to generation without change, except for rare mutational events. Over time, genetically isolated populations develop unique DNA mutations that are specific for that population group. These groups are also known as haplogroups.

    Y Chromosome Haplogroups

    The Y-chromosome is an extremely powerful indicator of ethnic origin. The lack of recombination within the Y chromosome has resulted in the chronological accumulation of ancient DNA point mutations, or single nucleotide polymorphisms (SNPs), that are associated with population development and the peopling of the world. The entire male population can be classified into 18 major haplogroups, A through R. Additional mutations within each of the major haplogroups further define subclades that typically illustrate more localized distribution reflecting recent human migrations.

    Mitochondrial Haplogroups

    Mitochondrial DNA is similarly a strong indicator of ethnic origin, although it is not as strong an indicator as the Y chromosome. While the reason is unclear, perhaps this is a result of the historical tendency for males to settle closer to their birthplaces than females.

    Worldwide, there are approximately 30 major mitochondrial haplogroups. These major haplogroups can be subdivided into sub-lineages which are often regionally or population specific.

    Limitations

    With over 500 ancestral origins reported in the 2000 U.S. Census, the United States is often characterized as a cultural melting pot. The 2000 census was the first to allow respondents to select more than one race. Nationwide, approximately 2.4 percent of the U.S. population, over 6.8 million Americans, marked identification with two or more races.

    European-American and Asian-American male populations demonstrate relatively low interracial admixture. In sharp contrast, the frequencies of European Y Chromosomes in Native American populations range widely, from 7-89%, depending on geographic location. In African American populations, the European Y Chromosome appears on average at a frequency of 26%. The Hispanic paternal gene pool tends to be a heterogeneous mixture of European, Native American and African Y Chromosomes.

    Summary

    The Y chromosome is the single most informative genetic marker for ethnic origin. While multi racial individuals can complicate interpretation of Y haplogroups, if used in conjunction with mitochondrial haplogroups, a broader view of a suspect’s ethnic origin can be estimated. Regardless, individuals with mixed ancestral background may not always display the expected physical characteristics. Consequently, results should always be interpreted with some degree of caution.

    US Paternity Laws can trace their origins to 18th Century England. Chief Justice of the King’s
    Bench, Lord William Murray Mansfield, served from 1756 until his retirement at age 80 in 1788. His
    decisions greatly influenced English common law, which remains the basis of law in the United
    States today. Sometimes referred to either as the Bastardy Statute or British common law origin,
    the Lord Mansfield’s Rule, defines a child born into a marriage to be a product of that marriage.
    The husband of the mother is legally bound to support the child as though it were his own. While
    use of the Mansfield Rule is falling on disfavor, many states still use this guiding principle. A

    Establishing Paternity

    Establishing paternity means being declared the legal father after the child is born. If the parents of
    a child were not married when the mother became pregnant or when the child was born, the child
    does not have a legal father until paternity is established.

    Establishing paternity is important for both the child and the parents. A child is entitled to the sense
    of belonging and identity that comes from knowing both parents. Establishing paternity often has an
    important emotional and psychological benefit for the child. Also, until there is a determination that
    a biological father is the legal father, the child has no right to receive financial support from the
    father, to inherit from the father or to obtain insurance, veterans’, social security or other benefits
    through the father. Likewise, until paternity is legally established, the biological father has no legal
    rights to the child, such as parenting time with the child. Additionally, he has no legal right to
    participate in major decisions about the child, such as medical treatment, education or religious
    training. Additionally, genetic diseases are inherited, thus knowing your biological ancestry can be
    useful in proactive care and transplantation. Paternity laws tend to vary from state to state. Click

    DNA results often decide the outcome of the case. It is important to understand the bottom line
    results on the paternity test report. All paternity test results and conclusions are founded in
    statistics and probabilities. The greater the number of genetic markers examined the greater the
    strength of the genetic evidence and hence the final result. Industry wide, paternity tests can be
    generated using as few as four markers, to as many as 16 markers, with the latest high resolution

    DNA Testing for Immigration Purposes

    There are a number of mechanisms for lawful entry into the United States. One popular method is for a citizen or lawful permanent resident of the United States to petition for an alien relative with form I-130. United States citizens can petition for their spouses, parents, children or siblings to immigrate to the United States and permanent residents can do so for their spouses and unmarried children.

    Normally the applicant uses a combination of birth, marriage, medical and other documents to prove that the applicant and the alleged relative are blood relatives.     New Florida Law Protects Men Falsely Accused of Paternity

    In June of this year Florida passed HB 7151 into law. The new bill permits a petition to set aside determination of paternity or terminate a child support obligation by an alleged father who has proven, through a DNA test that he is not the biological father of a child in the state of Florida.

    Previously, under Florida law, a man was required to pay child support until the child’s 18th birthday if it had been legally established that he was the father even if a DNA test at a later date proved he was not the biological father.

    The DNA Testing Technology:
